Description: Hela cells stably expressing Histone-2B fused to EGFP during cell division. Chromosome condensation as well as chromatin distribution during and after mitosis are cearly to be seen. Cells were grown on 35 mm glass bottom petri dishes.

Technical details: The 2-channel time lapse series was acquired on a Cell Observer system consisting of a fully motorized Axiovert 200M microscope, 40x Plan-Neofluar (NA=1.3) oil objective, Axiocam HRm and AxioVision 3.1 using the Multichannel- and Timelapse modules. Temperature control was achieved using the Incubator XL-3, Heating Insert P plus CO2 cover HM, providing a humidified atmosphere with 5% CO2. Imaging was done for 2 ½ hours with a lapse time of 5 minutes. Fluorescence illumination intensity was reduced to 1.5% using a neutral density filter employed in a motorized Filterwheel to prevent photodamage.
